Production of autoreactive I region-restricted T cell hybridomas
نویسندگان
چکیده
We produced autoreactive T cell hybridomas that can be induced to secrete interleukin 2 by co-culture with either syngeneic splenic stimulator cells or syngeneic Ia-positive B cell-B lymphoma hybridomas. These autoreactive hybridomas arose from the fusion of pork insulin-primed lymph node T cells with the AKR thymoma BW 5147 and occurred at a higher frequency than the expected insulin-specific hybridomas. Mapping studies using recombinant strains and blocking studies using monoclonal anti-Ia antibodies localized the stimulatory determinant to the I-Ad subregion of the major histocompatibility complex. These T cell hybridomas did not appear to be directed at any foreign antigen present in the culture system because activation occurred in serum-free, insulin-free medium (Iscove's medium). Such hybridomas should prove to be a potent tool in studying the biologic significance and function of the autoreactive response.

The role of Ia molecules in the activation of T lymphocytes. III. Antigen-specific, Ia-restricted, interleukin 2-producing T cell hybridomas with detectable affinity for the restricting I-A molecule
Antigen-specific I region-restricted, interleukin 2-producing T cell hybridomas were produced by fusing GAT-specific T cell blasts with BW5147. Two antigen-specific phenotypes were identified, one autoreactive and one nonautoreactive. All of the antigen-specific and autoreactive clones were H-2 restricted, mapping to the IA subregion by genetic analysis and monoclonal antibody inhibition. Both ...

The interaction of nominal Ag with major histocompatibility complex (MHC)-restricted T cells and accessory cells was studied by analyzing the effect of structurally related antigens on the response of antigen-specific MHC-restricted T cell hybridomas. The copolymer L-glutamic acid50-L-tyrosine50 (GT) completely inhibits the response of L-glutamic acid60-L-alanine40-L-tyrosine10 (GAT)-specific, ...
متن کاملA receptor presentation hypothesis for T cell help that recruits autoreactive B cells.
To uncover mechanisms that drive spontaneous expansions of autoreactive B cells in systemic lupus erythematosus, we analyzed somatic mutations in variable region genes expressed by a panel of (NZB x SWR)F(1) hybridomas representing a large, spontaneously arising clone with specificity for chromatin. A single mutation within the Jkappa intron that was shared by all members of the lineage indicat...
متن کاملAntigen-specific, H-2-restricted helper T cell hybridomas
We have examined the carrier-specific helper activity of a number of antigen-specific, I region-restricted T cell hybridomas prepared in our laboratory. The hybridomas were assayed for helper activity in the presence or absence of exogenously added nonspecific factors found in the concanavalin A-activated supernatants of normal mouse spleen cells. Of six hybridomas tested, all six could stimula...
متن کاملSerologic and functional characterization of a panel of antigen- presenting cell lines expressing mutant I-A class II molecules
An improved method is described for selecting mutant cells with an altered pattern of Ia antigenic determinants and antigen-presenting properties from an homogeneous population of functional antigen-presenting cells (APC). The APC line used, TA3, was a somatic cell hybrid obtained by fusing normal heterozygous H-2a/d B cells with a drug-marked variant of a BALB/c B lymphoma line. Two phenotypic...
